Officers arriving at the scene found two heavily damaged cars - a Hyundai Elantra and a Toyota Highlander.
The driver of the Hyundai was pronounced dead at the scene.
The two passengers of the Toyota were transported to a hospital with serious injuries.
Route 304 was temporarily closed from East Philips Hill Road to Squadron Blvd in both directions.
